Импорт тРНК в митохондрии дрожжей: роль предшественника митохондриальной лизил-тРНК-синтетазы и функция импортируемой тРНК в митохондриальном матриксе (реферат, курсовая, диплом, контрольная)
Митохондрии — это многофункциональные органеллы эукариотических клеток, обеспечивающие клетки энергией за счет окислительного фосфорилирования, участвующие в образовании железосерных кластеров, окислении жирных кислот, образовании некоторых аминокислот, а также играющие важную роль в апоптозе. Для митохондрий характерно наличие собственной ДНК и системы биосинтеза белка, однако большое число макромолекул импортируется в органеллы из цитоплазмы. Было показано, что в ряде случаев в митохондрии транспортируются не только белки, но также и тРНК. К настоящему времени импорт тРНК в митохондрии был обнаружен у простейших, ряда растений, сумчатых млекопитающих, а также у почкующихся дрожжей Saccharomyces cerevisiae. Механизм переноса тРНК в митохондрии и его специфичность значительно отличается от вида к виду.
В случае дрожжей было показано, что из цитоплазмы в митохондрии импортируется два вида тРНК — тРНКЛизСии (Martin et al. 1979) и тРНКГлн (Rinehart et al. 2005). Детальные механизмы импорта тРНК, а также функции импортируемой тРНК в митохондриалыюм матриксе, остаются неясными. Импорт тРНК является высокоспецифичным процессом, так как одна из двух изоакцепторных лизиновых тРНК (тРНКЛизсии, далее тРЛ1) направляется в митохондрии дрожжей, в то время как вторая (тРНКЛизиии> тРЛ2) присутствует только в цитоплазме. Такое различие в локализации довольно близких тРНК может быть объяснено отличиями в структуре этих тРНК (Entelis et al. 1998). Помимо этого, в нашей лаборатории было показано участие трех растворимых цитоплазматических белков в импорте тРЛ1 в митохондрии. Прежде всего, тРЛ1 должна быть аминоацилирована с помощью цитоплазматической лизил-тРНК-синтетазы, затем необходимо ее взаимодействие с предшественником митохондриалыюй лизил-тРНК-синтетазы (preMsklp) (Tarassov et al. 1995b). Предполагается, что тРЛ1 импортируется в митохондрии, будучи связанной с этим белком. Однако присутствия аминоацилированной тРЛ1 и preMsklp не достаточно для импорта тРЛ1 в митохондрии, что говорит о необходимости дополнительных белковых факторов, направляющих ее импорт, так называемых факторов импорта. Один из таких факторов был недавно охарактеризован как гликолитический фермент енолаза (Entelis et al. 2006). Роль этого белка в процессе импорта тРНК в митохондрии остается неясной.
Вопрос о функции tPJII в митохондриальном матриксе к настоящему моменту остается открытым. Существует предположение, что тРЛ1 принимает участие в митохоидриальной трансляции. Однако, эта гипотеза подтверждена только косвенными экспериментами (Kolesnikova et al. 2000). Более того, митохондриальная лизиновая тРНК дрожжей (с антикодоном cmnmsUUU, далее тРЛЗ) способна узнавать оба лизиновых кодона (AAA и AAG) в процессе митохондриалыюй трансляции.
Целью данной работы являлось изучение формирования комплекса между тРЛ1 и preMsklp, и с использованием полученных данных, создание штамма дрожжей, в котором тРЛ1 присутствовала бы исключительно в цитоплазме, и изучение фенотипического эффекта отсутствия тРЛ1 в митохондриях.
Для достижения поставленных целей были сформулированы следующие экспериментальные задачи:
— оценить константу диссоциации комплекса тРЛ1-preMsklp в отсутствии и присутствии енолазы 2;
— определить, какие нуклеотиды тРЛ1 становятся защищенными от действия нуклеаз в результате взаимодействия с preMsklp;
— оцепить эффективность импорта тРЛ1, направляемого различными мутантными версиями preMsklp in vitro и in vivo;
— в случае невозможности направления импорта тРЛ1 той или иной мутантной версией preMsklp, оценить эффективность митохондриального дыхания и митохондриалыюй трансляции в клетках штамма дрожжей с соответствующей мутантной версией preMsklp;
— оценить эффективность митохондриального дыхания и митохондриалыюй трансляции в клетках штамма дрожжей, в которых геи preMsklp заменен на ортологичный ген грибов Ashbya gossypii.
5. ВЫВОДЫ.
1. Только те тРНК, которые способны образовывать стабильный комплекс с предшественником митохондриалыюй лизил-тРНК-синтетазы (preMsklp), импортируются в изолированные митохондрии дрожжей. Константа диссоциации таких комплексов составляет 180−250 нМ. Присутствие енолазы снижает константу диссоциации до 40 нМ.
2. Комплекс импортируемой лизиновой тРНК (тРЛ1) с цитоплазматической лизил-тРНК-синтетазой организован как типичный комплекс аминоацил-тРНК-синтетазы класса НЬ с соответствующей тРНК, образование которого ведет к аминоацилированию. Комплекс тРЛ1 с preMsklp организован совершенно иначе: в этом комплексе подавляющее большинство нуклеотидов тРНК экранированы белком.
3. N-концевой домен preMsklp специфически направляет импорт тРНК в митохондрии дрожжей in vitro и in vivo и взаимодействует с тРЛ1 таким же образом, что и полноразмерный белок.
Участок preMsklp, определяющий специфический импорт тРНК в митохондрии, ограничен a-спиралями Н5 и Н6. Участки preMsklp, ограниченные a-спиралями Н5 и Н6 и a-спиралями HI и НЗ, необходимы для аминоацилирования зрелым белком митохондриальной лизиновой тРНК.
4. тРЛ1 участвует в митохондриальной трансляции. При 30 °C митохондриальная лизиповая тРНК (с антикодоном UUU) полностью заменяет в этом процессе тРЛ1 (с антикодоном CUU), тогда как при 37 °C митохондриальная лизиновая тРНК гипомодифицирована по U34 и неэффективно считывает кодоны AAG. Поэтому при данной температуре тРЛ1 необходима для нормального функционирования митохондрий.
