Структурно-функциональный анализ генов, кодирующих специфические субъединицы РНК-полимераз II и III делящихся дрожжей: Schizosaccharomyces pombe (реферат, курсовая, диплом, контрольная)
Транскрипцию ядерных генов в эукариотической клетке осуществляют три формы ДНК-зависимых РНК-полимераз, каждая из которых специфична в отношении считываемых ею генов. РНК-полимераза I (или А) считывает кластер генов рРНК, и продуктом этого процесса является предшественник высокомолекулярной рибосомной РНК. РНК-полимераза П (или В) синтезирует все мРНК и многие малые ядерные РНК (мяРНК), такие как U1 и U2. Продуктами генов, транскрибируемых РНК-полимеразой Ш (или С), являются низкомолекулярные РНК, участвующие в синтезе белков (тРНК и 5S РНК), их внутриклеточном транспорте (7SL РНК), сплайсинге мРНК (U6 РНК), посттранскрипционном процессинге рРНК и тРНК (соответственно MRP и HI РНК).
Впервые существование трех форм ДНК-зависимых РНК-полимераз было продемонстрировано Р. Рёдером и В. Рутером [1] более тридцати лет назад. Впоследствии три вида РНК-полимераз были выделены из ядер многих эукариотеческих организмов и стало очевидным, что РНК-полимеразы от дрожжей до многоклеточных обладают поразительно консервативным планом строения и содержат от 12 до 17 субъединиц [2,3].
Соотношение РНК к ДНК в быстро растущей клетке Saccharomyces cerevisiae составляет 50:1, при этом система транскрипции с участием РНК-полимеразы I является самой активной: доля рРНК составляет 80% от общего количества клеточных РНК, 15% ядерных транскриптов представлено молекулами тРНК, пул мРНК составляет лишь 5%. В отличие от РНК-полимераз П и Ш, находящихся в нуклеоплазме, РНК-полимераза I находится в ядрышке.
К данному моменту наиболее охарактеризованным объектом с точки зрения понимания механизмов транскрипции являются пекарские дрожжи S. cerevisiae [2−4]. Делящиеся дрожжи Shizosaccharomyces pombe, также принадлежащие к ветви аскомицетов, но разошедшиеся в эволюции с S. cerevisiae около миллиарда лет назад [5], представляют собой интересную модель для изучения процесса транскрипции в эукариотической клетке, поскольку по принципиальной организации некоторых биологических процессов (механизм сплайсинга, строение центромер, клеточный цикл) являются системой более приближенной к клеткам высших эукариот, чем S. cerevisiae.
Изучение ядерных РНК-полимераз дрожжей Sch. pombe было начато в нашей группе в рамках проекта по установлению структурно-функциональной консервативности базового аппарата синтеза РНК у эукариот. Вначале были клонированы кДНК и гены, кодирующие 7 общих субъединиц (Rpb5, Rpb6, Rpb8, RpblO, RpclO, Rpcl9 и Rpc40) ядерных РНК-полимераз I-Ш Sch. pombe, и проведены опыты по изучению их способности функционировать в клетках почкующихся дрожжей S. cerevisiae [6−10]. В 2001 году был завершен сравнительный анализ специфических субъединиц РНК-полимеразы I этого организма [11].
Данная диссертация является частью проекта, проводимого в группе механизмов генной экспрессии ИБХ РАН, по характеристике всех компонентов ядерных ДНК-зависимых РНК-полимераз I-Ш делящихся дрожжей Sch. pombe и посвящена структурно-функциональному анализу кДНК и генов, кодирующих специфические субъединицы РНК-полимераз II и Ш Sch. pombe. В работе особое внимание уделялось функциональному сравнению специфических субъединиц РНК-полимеразы Ш Sch. pombe и S. cerevisiae с тем, чтобы оценить консервативность и ход эволюции этого многосубъединичного фермента.
1. СТРУКТУРА И ФУНКЦИИ ТРАНСКРИПЦИОННОГО АППАРАТА ЭУКАРИОТИЧЕСКОЙ РНК-ПОЛИМЕРАЗЫ Ш.
Обзор литературы.
Данный обзор посвящен механизмам транскрипции, осуществляемой РНК-полимеразой Ш эукариот. Обобщены сведения о структуре промоторов, узнаваемых ядерной РНК-полимеразой Ш, рассмотрены специфические факторы транскрипции, механизмы инициации, элонгации и терминации процесса синтеза малых стабильных РНК в клетке, субъединичный состав РНК-полимеразы Ш и регуляция ее активности.
выводы.
1. Из кДНК-клонотеки Sch. pombe извлечена полноразмерная кДНК гена грЬ4+, функциональный анализ которой в клетках S. cerevisiae показал, что клонированная кДНК rpb4+ кодирует компонент РНК-полимеразы II — специфическую субъединицу Rpb4 Sch. pombe. С обнаружением субъединицы Rpb4 была завершена характеристика всех двенадцати компонентов РНК-полимеразы II Sch. pombe.
2. Установлена экзон-интронная структура гена rpbT, а также хромосомная локализация гена грЪ9кодирующих специфические субъединицы Rpb7 и Rpb9 РНК-полимеразы II Sch. pombe.
3. Два клона, pYUK71 и pYUG7, изолированных из геномной клонотеки Sch. pombe в процессе клонирования генов грЬ7* и грЬ9+ и использованных для гибридизации с космидными фильтрами клонотек Sch. pombe, позволили восстановить непрерывную нуклеотидную последовательность хромосомы I Sch. pombe в двух участках генома, для которых не было получено перекрывания соседних контиг.
4. Клонированы и секвенированы кДНК, кодирующие специфические субъединицы Rpcll, Rpcl7, Rpc25, Rpc31, Rpc34 и Rpc53 РНК-полимеразы III Sch. pombe, и тем самым завершена характеристика первичных структур малых специфических субъединиц РНК-полимеразы III делящихся дрожжей.
5. Функциональное тестирование в клетках S. cerevisiae восьми кДНК (rpclT, грс25+, грс31+, грс34+, грс53+, грс82грс2+ и rpct), кодирующих специфические субъединицы РНК-полимеразы III Sch. pombe, показало, что заменять in vivo ортологичные субъединицы почкующихся дрожжей способны только субъединицы Rpcl 7 и Rpc25 Sch. pombe, при этом клетки S. cerevisiae приобретают термочувствительный фенотип.
6. Получен активный препарат РНК-полимеразы III Sch. pombe, что позволяет расширить круг биохимических исследований этого фермента.
Заключение
.
В последнее десятилетие достигнут значительный прогресс в понимании фундаментальных аспектов транскрипции генов эукариотических организмов.
Изучение белок-белковых контактов и контактов белок-ДНК позволило понять процесс формирования инициаторных комплексов на различных типах промоторов РНК-полимеразы 1П. Накоплен большой объем знаний о взаимодействии субъединиц в составе фермента. Однако до сих пор нет детального понимания того, что происходит внутри элонгационного комплекса при попадании его в сайты терминации, что вызывает высвобождение РНК-транскрипта и диссоциацию РНК-полимеразы с матрицы. Нет подробной динамической модели процесса транскрипции эукариотических генов от инициации до терминации. Ответы на эти вопросы предстоит найти в ближайшем будущем.
