Синтез и свойства хромофоров белков семейства зеленого флуоресцентного белка (реферат, курсовая, диплом, контрольная)
Бурно развивающиеся в последние годы технологии флуоресцентной микроскопии позволили визуализировать многие биологические процессы. Одним из важнейших инструментов изучения живой клетки являются флуоресцентные маркеры, дающие возможность проводить наблюдение за целевыми структурами или процессами в режиме реального времени. Особенно широко используются генетически кодируемые метки — флуоресцентные белки семейства зеленого флуоресцентного белка GFP, формирующие хромофорную группу внутри молекулы белка за счет посттрансляционных модификаций собственных аминокислотных остатков. Исследования последних лет показали большое разнообразие спектральных характеристик и структур хромофоров в GFP-подобных белках.
Данная работа посвящена синтезу и изучению химических и. спектральных свойств хромофорных групп GFP-подобных белков. Впервые разработаны методы синтеза хромофоров флуоресцентных и хромобелков — гомологов GFP, поглощающих и флуоресцирующих в красной области видимого спектра. Синтезированы хромофоры пурпурного хромобелка asFP595, красного фотоконвертируемого флуоресцентного белка Kaede и желтого флуоресцентного белка zFP538. Синтетические хромофоры позволили провести детальный анализ ауксохромных свойств заместителей в данном ряду окрашенных соединений, а также их сольватохромных и кислотно-основных свойств. Выявлена химическая природа сложного спектрального поведения хромопептидов из белка zFP538, приведшего к неправильному определению их структуры. Обнаружена необычная перегруппировка 2-ацил-5-арилиден-3,5-дигидро-4//-имидазол-4-онов в уникальные производные 2,6-дикетопиперазина, встречающиеся в природе среди метаболитов грибов. Полученные в настоящей работе флуорогены (красители, практически нефлуоресцентные в свободном состоянии в растворе, но приобретающие яркую флуоресценцию после прочного связывания с целевой молекулой, например, белком или ионом металла) и их аналоги могут найти применение для различных флуоресцентных методов детекции.
ГЛАВА 1. Биохимия флуоресцентных белков и их хромофоров (обзор литературы).
1.1 Введение.
Белки семейства зеленого флуоресцентного белка GFP широко известны благодаря уникальному механизму формирования хромофорной группы, а также благодаря возможности их использования в качестве генетически кодируемых флуоресцентных меток для прижизненной визуализации процессов жизнедеятельности отдельных клеток и целых организмов [1, 2]. Отличительной особенностью белков этого семейства является уникальный автокаталитический механизм формирования хромофора. В отличие от обычных ферментов, производящих какую-либо одну реакцию множество раз, флуоресцентные белки в ходе своего созревания один раз проводят 2−3 совершенно разные реакции, приводящих к синтезу хромофора внутри молекулы белка за счет посттрансляционных модификаций собственных аминокислотных остатков.
Первый зеленый флуоресцентный белок был найден в гидроидной медузе Aequorea victoria [3, 4]. Позже в коралловых полипах были открыты желтые, оранжевые и красные флуоресцентные белки, а также нефлуоресцентные хромобелки [2, 5]. Исследования последних лет показали, что спектральное разнообразие белков семейства GFP обеспечивается разнообразием химических структур их хромофоров [6, 7].
Синтез модельных хромофоров флуоресцентных белков тесно связан с проблемой определения химического строения нативных хромофоров.
Доказательство строения GFP-хромофора проходило в несколько этапов.
Вначале первооткрывателем GFP Шимомурой была установлена аминокислотная последовательность пептида, полученного при 6 гидрировании окрашенной фракции лизата GFP и выделен п-гидроксибензальдегид в качестве продукта гидролиза хромофора, показано спектральное сходство синтезированного de novo хромофора и окрашенного продукта протеолиза GFP [8]. Значительно позже, после успешного клонирования гена GFP [4], было проведено сравнение структур белкового и синтетического хромофоров методом масс-спектрометрии [9], а также непосредственное определение структуры нативного белка с помощью рентгеноструктурного анализа (РСА) [10, 11]. В настоящее время PC, А стал основным методом исследования структуры хромофоров в различных флуоресцентных белках [7]. В то же время, кристаллографическое определение структуры не всегда допускает однозначную интерпретацию и малоинформативно при изучении химии и фотохимии хромофоров. Именно поэтому синтез и изучение низкомолекулярных модельных соединений остаются важными задачами, особенно ввиду возрастающего разнообразия известных структур хромофоров.
Несмотря на то, что ковалентная структура большинства известных белковых хромофоров считается надежно установленной, детали стереохимии, кислотно-основных взаимодействий, а также реакционной способности возбужденного состояния остаются недостаточно изученными. В то же время, перечисленные особенности хромофоров важны для понимания таких процессов, происходящих в нативных белках, как фотоактивация [12], фотопереключение [13], фотообесцвечивание [14], фототоксичность [15], созревание сложных хромофоров и др.
В данном обзоре рассмотрены структурные и биохимические особенности флуоресцентных белков — гомологов GFP, методы синтеза соединений, моделирующих хромофоры GFP-подобных белков, обобщены результаты, полученные при изучении этих соединений, и их использование для объяснения спектральных и биохимических свойств флуоресцентных белков.
1.2 Флуоресцентные белки — гомологи зеленого флуоресцентного белка (GFP).
Выводы.
1. Разработан общий метод синтеза 5-арилиден-3,5-дигидро-4//имидазол-4-онов, имеющих ацильный заместитель в положении 2 имидазола. С использованием этого метода получен хромофор окрашенного белка asFP595, изучено его спектрохимическое поведение в различных условиях. Показана тождественность структуры модельного соединения структуре хромофора в белке.
2. Разработан новый метод синтеза 5-арилиден-3,5-дигидро-4//-имидазол.
4-онов, имеющих функционализированный этенильный заместитель в положении 2 имидазола. С использованием этого метода получен хромофор флуоресцентного фотопереключаемого белка Kaede (красная форма). Также синтезированы б структурных аналогов хромофора Kaede, соответствующих замене гистидина на другие аминокислоты в хромофоре. Изучены спектрохимические характеристики полученных хромофоров. Показано, что хромофор Каеёе-типа, содержащий остаток аспарагина вместо гистидина обладает наиболее длинноволновым поглощением.
3. Получен хромофор желтого флуоресцентного белка zFP538.
Независимо показано, что в нативном zFP538 хромофор существует в форме внутримолекулярного имина. Выявлена химическая природа сложного рН-зависимого спектрального поведения хромофора ZFP538.
4. Обнаружена самопроизвольная перегруппировка 5-арилиден-2-ацил.
3,5-дигидроимидазол-4-онов в 3-арилиден-5-алкил-2,6дикетопиперазины в водных и спиртовых растворах в кислой среде.
