Изучение регуляторной функции С-концевого сегмента рековерина (реферат, курсовая, диплом, контрольная)
Ключевые аспекты функционирования нейронов регулируются за счет изменений концентрации внутриклеточного кальция, которые в зависимости от своей интенсивности и продолжительности активируют различные сигнальные каскады, приводящие к конкретным физиологическим эффектам. Разнообразие этих эффектов является результатом действия белков семейства нейрональных кальциевых сенсоров (НКС), способных распознавать сигналы кальция и преобразовывать их в широкий спектр клеточных ответов. За счет этой способности ИКС вовлечены в сигнальные механизмы, обеспечивающие нормальную жизнедеятельность нейронов, начиная от роста и выживаемости, и заканчивая сенсорной функцией, нейротрансмиссией и всеми этапами синаптической пластичности. Более того, нарушение экспрессии и (или) функционирования НКС приводит к активации патогенетических сигнальных механизмов, ассоциированных с целым рядом заболеваний центральной нервной системы.
Примером сигнального каскада с участием НКС является передача зрительного сигнала в фоторецепторной клетке (фототрансдукция). Центральным элементом Са2+ -зависимой регуляции фототрансдукции является НКС рековерин, функция которого заключается в придании процессу де-сенситизации зрительного рецептора родопсина чувствительности к ионам кальция. При высокой концентрации кальция, характерной для темнового состояния фоторецепторной клетки, рековерин ингибирует фосфорилиро-вание родопсина, катализируемое родопсинкиназой. В ответ на световой стимул происходит снижение концентрации кальция в цитоплазме фоторецепторной клетки, рековерин теряет способность ингибировать родопсин-киназу, в результате чего происходит фосфорилирование родопсина, инициирующее быстрое выключение фотовозбужденного рецептора.
Белки семейства НКС, включая рековерин, обладают высокой степенью структурного сходства. Гомология распространяется на структуру их.

Несмотря на интенсивные исследования НКС, проводимые в последнее время, взаимосвязь между структурной организацией этих белков и специфичностью их регулятор-ной активности до сих пор остается неустановленной. Кроме того, обнаружение новых мишеней для НКС усложняет решение этой проблемы. В связи с этим актуальной задачей является поиск регуляторных элементов в структуре НКС, которые могли бы обеспечивать уникальность функциональных свойств каждого из этих белков. Мы предположили, что одним из таких элементов может быть С-концевой сегмент — последовательность, которая среди НКС отличается вариабельностью структуры всех уровней. В настоящей работе исследована роль С-концевого сегмента в обеспечении эффективности и специфичности Сазависимого функционирования НКС рековерина.

выводы.
1. Удаление участка С-концевого сегмента рековерина, состоящего из 14-ти С-концевых аминокислотных остатков, не приводит к нарушению целостности структуры белка.
2. Аминокислотные остатки 191(ЗКУКЕК196 в С-концевом сегменте рековерина принимают участие в настройке его чувствительности к ионам кальция и, как следствие, в регуляции Са2±миристоильного переключателя белка.
3. Образование контактов между С-концевым сегментом рековерина и 14-концевым 15-звенным участком родопсинкиназы, включая катион-тс взаимодействие К192кс-Р3кк, стабилизирует связывание родопсинкиназы с гидрофобным карманом рековерина, тем самым обеспечивая эффективное ингибирование фермента.
4. Замена С-концевого сегмента рековерина на соответствующую последовательность другого нейронального кальциевого сенсора, вСАР2, ослабляет ингибирование родопсинкиназы рековерином.
