Изучение взаимодействия TFIID — общего фактора транскрипции и PBAP — комплекса, ремоделирующего хроматин (реферат, курсовая, диплом, контрольная)
Живые организмы — сложно организованные системы, способные хранить и передавать потомкам свою генетическую информацию. Принципы и механизмы реализации наследственной информации интересуют исследователей с момента открытия основного постулата молекулярной биологии (ДНК О РНК=>бел ок). Генетическая информация о структуре отдельных белков и нуклеиновых кислот у всех организмов заключена в молекулах ДНК или РНК в виде последовательностей нуклеотидов, называемых генами. Координированная работа (экспрессия) большого числа генов возможна лишь благодаря наличию тонких регуляторных механизмов, определяющих место, время и уровень экспрессии конкретного гена или группы генов.
Экспрессия генов в эукариотической клетке представляет собой сложный многостадийный процесс, включающий. в себя: транскрипцию генов (переписывание генетической информации с ДНК на специальные матричные РНК (мРНК)), процессинг и сплайсинг мРНК (модификации мРНК и удаление избыточных частей последовательностей мРНК), экспорт упакованных мРНК из ядра в цитоплазму, трансляцию (построение на основе мРНК белковой последовательности) и посттрансляционные модификации белков. Важнейшим этапом является транскрипция, осуществляемая главным ферментом ДНК-зависимой РНК-полимеразой и многочисленными регуляторными белками — факторами транскрипции. Факторы транскрипции взаимодействуют с регуляторными нуклеотидными последовательностями генов, друг с другом, с молекулами РНК-полимеразы и необходимы для правильного узнавания транскрипционным комплексом регуляторных последовательностей в составе генов, а их координированная работа приводит к повышению или понижению уровня или активации транскрипции соответствующих генов при ответе клеток на внешние или внутренние регуляторные сигналы.
В эукариотической клетке ДНК всегда связана с белками и организована в хроматин, который находится в различных состояниях, отличающихся степенью компактизации и транскрипционной активностью. Степень компактизации хроматина определяет доступность регуляторных элементов действию транскрипционных факторов, а соответственно, и транскрипционную активность. Существуют разнообразные коактиваторы транскрипции, которые в ответ на действие активаторов способны изменять строение хроматина и структурировать ДНК-матрицу для инициации транскрипции и функционирования транскрипционного комплекса. Одними из коактиваторов транскрипции являются хроматин-ремоделирующие комплексы.
ОБЗОР ЛИТЕРАТУРЫ.
выводы.
1. Выделен и охарактеризован новый комплекс, состоящий из общего фактора транскрипции TFIID, ремоделирующего хроматин комплекса РВАР и коактиватора транскрипции SAYP. Показано, что в состав нового комплекса входят все субъединицы комплексов TFIID и РВАР.
2. SAYP — основной фактор клетки, объединяющий TFIID — общий фактор транскрипции и РВАР — комплекс, ремоделирующий хроматин.
3. Единый комплекс, содержащий РВАР, TFIID и SAYP, локализуется на промоторах SAYP-зависимых генов и необходим для рекрутирования РНК-полимеразы II.
4. Все компоненты общего комплекса (SAYP, TFIID, РВАР) необходимы для его эффективного связывания с промоторами SAYP-зависимых генов.
