Создание первой карты генома Aktinobacillus pleuropneumonie и сравнительное макрорестрикционное картирование 12 референтных штаммов (реферат, курсовая, диплом, контрольная)
Actinobacillus pleuropneumoniae (A. pleuropneumoniae) — этиологический агент актинобациллезной (гемофиллезной) плевропневмонии свиней, инфекционной контагиозной болезни, которая характеризуется при остром течении геморрагическим воспалением легких и фибринозным плевритом, а при подостром и хроническом — развитием очаговой гнойой некротизирующей пневмонии и фибринозным плевритом. При хроническом течении в легких образуются секвестроподобные изменения, в которых у животных-реконвалесцентов возбудитель может сохраняться длительное время. Такие животные («carrier») наиболее опасны как источник возбудителя.
Изоляты A. pleuropneumoniae встречаются в 12 серологических вариантах, отличающихся не тоько по составу капсульного антигена и ли-пополисахарида клеточной стенки, но и по вирулентности. Различные серовары могут быть объединены в группы на основании профилей протеинов наружной мембраны, антигенного сходства, наличия токсинов или на основании полиморфизма длины рестрикционных фрагментов.
У A. pleuropneumoniae были описаны некоторые факторы вирулентности, такие как RTX-токсины Apxl, Apxll und Apxlll, трансферрин-связывающие протеины TbpB и TbpA, супероксиддисмутаза SodA, липо-протеины наружной мембраны OmIA и капсульные антигены. Несмотря на это, в настоящее время не существует вакцины, обеспечивающей перекрестный иммунитет.
Несмотря на детальную информацию о классификации серотипов и конкретных факторах вирулентности, данные о структуре генома A. pleuropneumoniae отсутствуют. Как показано на примере других бактерий, такая информация является важной предпосылкой как для детального понимания механизмов вирулентности и патогенеза, так и для создания эффективных средств профилактики. 10 генома может быть получена путем полного его секвенирования, либо путем построения карты генома.
Поскольку полное секвенирование генома A. pleuropneumoniae в обозримом будущем не представляется возможным, в рамках данной работы планируется создать первую карту генома A. pleuropneumoniae, используя методы гелевого пульс-электрофореза и гибридизации по Саузерн. Дополнительно планируется с использованием этих же методов выявить сходство и различия между референтными штаммами различных серотипов A. pleuropneumoniae. Результаты данной работы должны послужить базой для дальнейших более детальных исследований.
2 Обзор литературы.
4 Результаты исследований.
4.1 Физическая карта A. pleuropneumoniae АР76.
Содержание гуанина и цитозина (мол. ГЦ %) в геноме A. pleuropneumoniae составляет примерно 42% (MANNHEIM 1984). Поэтому, для получения относительно небольшого количества макрорестрикционных фрагментов, хорошо различимых по величине, мы расщепляли ДНК A. pleuropneumoniae рестрикционными эндонуклеазами, проявляющими активность в участках с преимущественным содержанием гуанина и цитозина. Были выбраны следующие эндонуклеазы: Ара (5'-ГГГЦЦ/Ц-3'), AscI (5'-ГГ/ЦГЦГЦЦ-3'), Not (5'-ГЦ/ГГЦЦГГ) и Sal (5'-Ц/ТЦГАЦ-3'). За исключением Sa/I выбранные энзимы расщепляли ДНК A. pleuropneumoniae на количество фрагментов, образующих после электрофо-ретического разделения хорошо анализируемый узор (рис. 2). Для энзима Sa/I был установлен единственный сайт узнавания, который мы взяли за исходный пункт («zero point») физической карты генома A. pleuropneumoniae.
Фрагменты получали название в зависимости от используемого энзима (АРА, ASC, NOT) и нумеровались соответственно относительного положения в геле (наибольший фрагмент — 1, второй по величине — 2 и т. д.). Всего было найдено 6 фрагментов для энзима AscI, 8 для Not и 11 для Ара. Размеры фрагментов представлены в таблице 6.
