Теоретическое обоснование и практическое использование молекулярно-генетических методов в защите сельскохозяйственных растений от вредителей и оценке трансгенных растений на биобезопасность (реферат, курсовая, диплом, контрольная)
Для успешного осуществления программ защиты сельскохозяйственных растений от вредных членистоногих необходимо изучение биологии и генетики популяций как вредных, так и полезных насекомых. Это включает в себя знание генетической структуры популяций, миграционных процессов (динамики), акклиматизации, поведенческих реакций, условий размножения, отношения полов и трофических связей.
Значительный прогресс в этом отношении был достигнут в 50−60-е годы прошлого столетия, благодаря использованию классических генетических принципов и подходов. Изучение популяционных процессов насекомых исследователи проводили, главным образом, с помощью видимых и хорошо различимых фенотипических маркеров (морфологических и фенетических), таких как окраска и рисунок, волоски или шипы на теле особи. Это приблизило ученых к пониманию закономерностей распространения насекомых, их поведенческих реакций, включая половые отношения, и наследования отдельных генов, контролирующих те или иные признаки [ВеЬига, 2006].
Использование молекулярно-генетических подходов, начиная с белковых маркеров в 1970;х годах, главным образом изоферментов (продуктов различных локусов, но со сходной функцией) и позже ДНК-маркеров, во многом способствовало более глубокому пониманию исследуемых закономерностей в популяциях насекомых. Большинство ДНК-маркеров, используемых сегодня — это продукты полимеразной цепной реакции (ПЦР), которые могут быть получены из экстремально малого количества биоматериала (из яйца, личинки, куколки, имаго насекомого или его отдельных органов).
В последние несколько лет использование молекулярных маркеров в энтомологии широко обсуждалось в различных обзорах [Удалов и др., 2003;
Behura, 2006.,' Loxdale, MacDonald, 2004; MacDonald., Loxdale, 2004]. Молекулярные маркеры позволяют анализировать и объяснять популяционные процессы там, где этого не могут сделать никакие другие методы исследований. Использование ДНК маркеров необходимо для анализа структуры популяций как полезных насекомых — паразитов и хищников — так и вредителей, являющихся объектом контролирующих их биоагентов. Кроме того, они могут использоваться для целей таксономии и филогении [Roehrdanz, Flanders, 1993; Mitchell et al., 2006]. С их помощью можно разделить таксоны насекомых, т. е. биотипы, подвиды, близкородственные виды, а также виды-двойники, т. е. виды, которые трудно различить морфологически или каким-то другим способом [Mitchell et al., 2005, Mitchell, Samways, 2005; Silva-Brandao et al., 2008].
Несмотря на столь широкие возможности ДНК-маркеров, сегодня в нашей стране в генетических исследованиях популяций вредителей преобладает использование морфологических и фенетических маркеров. На их основе исследователи продолжают изучать динамику, поведенческие реакции и строят прогнозы о развитии резистентности насекомых к инсектицидам, главным образом для сельскохозяйственно-значимых вредителей [Фасулати, Вилкова, 2000; Сухорученко, 2001, 2005; Король, Новосельская, 2001; Беньковская и др, 2004; Ростовцева, 2005;].
Несмотря на то, что фенотипические маркеры просты для использования и часто проявляются на протяжении всего жизненного цикла организма, они имеют ряд существенных недостатков. Основными ограничениями их использования является то, что хорошо различимые фены относительно редки и встречаются далеко не у всех видов насекомых. Проблема идентификации вида по морфологическим признакам в отдельных случаях значительно затрудняется из-за существования видов-двойников. Кроме того, модификационная изменчивость фенотипических маркеров, как правило, весьма значительна, что б затрудняет оценку и прогноз динамики популяционных процессов. Более того, идентификация таких маркеров должна базироваться на знании их генетического контроля и того как гены, контролирующие этот признак, наследуются в потомстве.
Использование для этих целей современных методов молекулярно-генетического анализа, в частности, ПЦР-метода и полученных на его основе ДНК-маркеров, может во многом способствовать решению этих проблем. Важно отметить, что использование ДНК-маркеров не умаляет применения фенетических и других морфологических критериев в практике защиты растений от вредителей, но лишь дополняет их и расширяет возможности для популяционных исследований видов насекомых, не имеющих четких видимых морфо-фенетических признаков, позволяет повысить точность мониторинга и прогноза. Кроме того, с использованием ДНК-маркеров появляется возможность проследить динамику отдельных генетических элементов, отдельных хромосомных локусов, генов и аллелей генов в популяциях, оценить гетерозиготность, гетерогенность и генетическое сходство популяций и другие параметры, которые невозможно оценить с помощью морфологических критериев.
Таким образом, сегодня исследователям недостает точных методов анализа и прогноза в популяциях вредных и полезных насекомых для целей мониторинга и защиты рстений. Также существует недостаток знаний о молекулярно-генетической структуре популяций насекомых и закономерностях ее изменчивости под влиянием стрессовых факторов внешней среды. В этой связи данные исследования, несомненно, актуальны и представляют интерес для практики защиты растений от вредных насекомых.
В то же время многие эксперты по сельскому хозяйству считают, что проблема нехватки продовольствия не может быть решена без применения ДНК технологий и, в частности, генной инженерии. Генетическая инженерия, по 7 сути, продолжает направление традиционной селекции по улучшению генотипов полезных растений, но достигает той же цели более эффективным и быстрым путем. На сегодняшний день генетическая инженерия уже располагает большим арсеналом знаний и методов для эффективного переноса полезных генов из одних организмов в другие [Романов, 2000]. На этой основе уже созданы многие сорта трансгенных или генетически-модифицированных растений (ГМР) и некоторые виды ГМ-насекомых, нашедшие сегодня применение в мировой практике защиты сельскохозяйственных растений. Однако их использование в нашей стране сдерживается недостаточной изученностью вопросов их экологической безопасности.
В этой связи данные исследования также актуальны и могут найти свое отражение в практике защиты растений на этапе предрегистрационных, регистрационных испытаний и пострегистрационного мониторинга ГМР и продукции на их основе.

В результате проведенных исследований можно сделать следующие основные выводы:
1. Созданы новые более информативные системы описания фенстической структуры популяций колорадского жука и клопа вредная черепашка.
2. Чувствительность различных фенетических групп насекомых к инсектицидам не является решающим фактором, определяющим фенооблик популяций колорадского жука. Фенетическая структура популяций колорадского жука и клопа вредная черепашка в первую очередь определяется соотношением внутрипопуляционных групп насекомых, имеющих различную неспецифическую устойчивость к различным стрессам, что указывает также на универсальный характер выявленных закономерностей.
3. Выявлена сцепленность генов окраски и рисунка имаго клопов вредной черепашки с полом. Изучение популяционных процессов клопа вредная черепашка по фенетическим маркерам необходимо проводить с учетом половых различий.
4. Выявлена высокая модификационная изменчивость фенетической структуры популяции клопа вредная черепашка, что ставит под вопрос использование фенетических маркеров при мониторинге резистентности клопов к инсектицидам.
9. Риск вертикального переноса генов от ^/-картофеля к его диким сородичам и культурным сортам минимален, тогда как риск вертикального переноса генов от ГМ-кукурузы к культурным сортам существует. Поля с трансгенными сортами кукурузы должны быть удалены не менее чем на 200 м от полей с культурными сортами, при этом необходимо создание барьеров на пути движения пыльцы.
10. Разработан метод полу количественной оценки содержания ГМИ, основанный на амплификации участка целевого гена СР4 ЕРБРБ с использованием пары праймеров: ЯШИ и ЫШ)4, фланкирующих участок целевого гена размером 356 п.н., который позволяет проводить не только ПЦР-детекцию и идентификацию трансгенов в зерне и зернопродуктах сои, но и определять их количественное содержание.
ПРАКТИЧЕСКИЕ РЕКОМЕНДАЦИИ.
1. Для целей фенетического анализа популяций колорадского жука и клопа вредная черепашка предлагается использовать новые системы описания феноформ рисунка и окраски имаго насекомых.
2. Для молекулярно-генетического анализа популяций насекомых предлагается использовать разработанную методику: «Методика оценки ДНК-полиморфизма популяций насекомых с помощью ПЦР (КАРБи ^БЯ-РСК)».
3. Поиск резистентных к инсектицидам генотипов в популяциях моновольтинных видов насекомых можно проводить с использованием предлагаемого нами подхода.
4. В целях быстрого и эффективного мониторинга ГМ-растений на полях и получаемых на их основе продуктов и кормов использовать предлагаемые нами методы детекции, идентификации и полуколичественной оценки трансгенной вставки в биоматериале.
5. Поля с трансгенными сортами кукурузы должны быть удалены не менее чем на 200 м от полей с культурными сортами, необходимо создание барьеров на пути движения пыльцы. Изоляции полей с трансгенным картофелем не требуется.
6. Мониторинг резистентности колорадского жука к трансгенному (В1:) картофелю проводить с использованием предлагаемого нами метода: «Метод оценки чувствительности популяций колорадского жука к трансгенному (В^ картофелю по феноформам рисунка переднеспинки имаго».
7. На этапе предрегистрационных испытаний оценку пролонгированного влияния трансгенных растений на нецелевых насекомых проводить с использованием предлагаемого нами подхода и, в частности, оценку В1-картофеля — по предлагаемой нами методике: «Методика оценки пролонгированного действия трансгенного (В1-) картофеля на нецелевых насекомых».
