Малые интерферирующие РНК Drosophila virilis и их роль в экспрессии мобильных элементов (реферат, курсовая, диплом, контрольная)
Геномы всех изученных организмов содержат в своём составе' мобильные генетические элементы (МЭ). Долгое время эту «мусорную» часть генома недооценивали и отводили ей пассивную роль в процессе эволюции. Однако, в настоящее время достоверно известно, что мобильные элементы являются неотъемлемой составляющей геномов и одной из движущих сил микроэволюции. С одной стороны, встраиваясь в промоторную или кодирующую части генов, они могут изменять их экспрессию или вызывать различные мутациис другой стороны, такие транспозоны как НеТ-А и ТАЯТ играют ключевую роль в поддержании длины теломерных участков хромосом у видов ВгозорЪИа.
Актуальность изучения МЭ не вызывает сомнения. Стоит упомянуть хотя бы открытие /'-элемента, с помощью которого стал возможен трансгенез И. melanogaster, получение разнообразных мутаций и открытие функций множества генов у этого вида. Исследование функций мобильных элементов и механизмов их контроля стало новой вехой в развитии молекулярной биологии и генетики на рубеже 20 и 21 веков. Особенно это стало актуальным в связи с открытием явления РНК-интерференции (РНКи), которое представляет собой совокупность высококонсервативных механизмов борьбы организма с экзогенными (вирусные инфекции) и эндогенными (мобильные элементы) нуклеиновыми кислотами. РНКи была впервые открыта у СаепогкаЬс1Ш. ь' е^апБ, однако уже скоро была доказана широкая распространённость данного явления среди множества эукариотических организмов, а её аналоги были найдены у прокариот и архей.
Особый интерес вызывает исследование механизмов, предотвращающих негативные последствия транспозиций МЭ в геноме терминальных клеток и, таким образом, способствующих нормальной передаче наследственной информации следующему поколению.
Следует подчеркнуть, что кроме РНК интерференции в ходе эволюции виды выработали и другие весьма эффективные механизмы, предотвращающие инвазии и перемещения МЭ в геномах. Поэтому большое значение для понимания механизмов, нейтрализующих вредное действие МЭ, имеет изучение случаев, при которых МЭ выходят из-под контроля и «распрыгиваются» по геному хозяина. К таким случаям относятся описанные у И. те1апо%а$ 1ег и В. утИх системы (ГД), когда при скрещивании линий, отличающихся по содержанию того или иного МЭ, данный элемент претерпевает массовые транспозиции в потомстве, что приводит к стерильности и другим нарушениям.
В целом, стоит отметить, что исследования общих генетико-молекулярных механизмов, обеспечивающих транспозиции различных МЭ, имеют большое значение для понимания роли МЭ в формировании структуры и контроле функционирования эукариотического генома.
1. ОБЗОР ЛИТЕРАТУРЫ.
ВЫВОДЫ:
1. Исследованы основные типы и характеристики малых интерферирующих РНК, гомологичных всему спектру мобильных генетических элементов генома D. virilis. Большинство транспозонов в геноме D. virilis являются мишенью одновременно для si-и piPHK путей в терминальных тканях.
2. Показано, что при искусственном введении МЭ Penelope в геном D. melanogasier, данный элемент процессируется только в 21-нуклеотидные siPHK, которые связаны с белком Ago2 — ключевым белком siPHK пути. Выявлена их преимущественная локализация в пределах инвертированных концевых повторов МЭ Penelope, которые могут образовывать двуцепочечные РНК-интермедиаты («шпильки») и являться мишенью siPHK пути.
3. Хотя ретротранспозон Ulysses и находится под контролем piPHK пути, но биогенез piPHK, гомологичных этому элементу, идет по обратному пинг-понг циклу, где смысловые транскрипты МЭ Ulysses являются источником первичных piPHK.
4. Среди обнаруженных классов Реие/оре-гомологичных коротких РНК, только piPHK передаются в потомстве и таким образом являются «материнским фактором», играющим важную роль в супрессии данного МЭ.
5. В геноме D. virilis описаны и локализованы piPHK локусы, проявляющие дифференциальную активность в разных линиях, а также впервые найдены множественные piPHK, гомологичные белок-кодирующим последовательностям.
6. Транскрипция МЭ Penelope начинается одновременно с началом общей транскрипции генома, спустя 2−5 часа с момента оплодотворения яйца, и предшествует гибели клеток зародышевого пути у эмбрионов от дисгенного скрещивания, что согласуется с важной ролью этого МЭ в дисгенезе.
