Дизайн, синтез и изучение связи структуры и анксиолитической активности N-ацилдипептидных аналогов холецистокинина-4 (реферат, курсовая, диплом, контрольная)
Состояние тревоги, переходящее в чувство страха и панику, возникает как у здоровых людей в результате эмоционального стресса, так и входит в симптоматику различных тревожно-фобических синдромов. Это различные невротические состояния с явлениями напряжения, беспокойства, повышенной раздражительности, тревоги и страха, тревожно-депрессивные состояния, нарушения сна, некоторые психосоматические заболевания (язвенная болезнь желудка и 12-ти перстной кишки) и др.
В настоящее время известно более 100 препаратов, способных в большей или меньшей степени регулировать данные состояния. Эти соединения, принадлежащие к классу психотропных средств и называемые анксиолитиками или транквилизаторами, относятся к разным химическим группам и обладают различными механизмами действия. Почти половину из них составляют препараты бензодиазепинового ряда, являющиеся прямыми агонистами ГАМК-бензодиазепинового рецепторного комплекса (диазепам, феназепам, клоназепам, лоразепам, оксазепам и др.). Помимо этого в отдельные группы выделяют анксиолитики, действующие через серотониновые (буспирон, закоприд и др.) и глутаматные рецепторы, а также ряд других (бенактезин, мебикар, афобазол и др.).
На сегодня наиболее широко применяемыми транквилизаторами остаются бензодиазепины, в первую очередь, благодаря своей ярко выраженной эффективности. Однако данный класс анксиолитиков обладает и рядом крайне нежелательных эффектов. Наиболее проблемными побочными эффектами являются развитие привыкания и формирование зависимости. Помимо этого, бензодиазепины оказывают седативное, миорелаксирующее, снотворное, амнестическое действие. Ряд этих эффектов позволяет использовать данные соединения в качестве противосудорожных или снотворных препаратов. Однако, в случае необходимости получения чистого противотревожного действия, данные проявления их активности оказываются крайне нежелательными побочными эффектами, сильно ограничивающими применение препаратов. В последнее время из-за такого обширного списка нежелательных эффектов были введены жёсткие меры по контролю за назначением и применением транквилизаторов бензодиазепинового ряда. В результате этого число зарегистрированных в России бензодиазепинов уменьшилось более чем на 50%. Поэтому создание новых анксиолитиков, чьё противотревожное действие будет осуществляться посредством взаимодействия с другим, не ГАМК-бензодиазепиновым рецепторным комплексом, и не проявляющих нежелательных побочных эффектов, на сегодняшний день актуально.
Существует достоверная информация об участии различных форм холецистокинина в регуляции паники и тревоги. В частности, известно об анксиогенных свойствах нейропептида холецистокинина-4 (ХЦК-4), селективно взаимодействующего с ХЦКг-типом рецепторов. На основании этих данных учёными разных стран было создано несколько селективных ХЦКг-антагонистов различной химической структуры, обладающих анксиолитическими свойствами, а также была показана возможность создания соединений с противотревожной активностью на базе структуры самого ХЦК-4. Однако ни один из полученных потенциальных анксиолитиков не прошёл стадию клинических исследований, и на сегодня среди транквилизаторов отсутствуют лиганды ХЦК-рецепторов, как пептидной, так и непептидной природы. Наиболее перспективным направлением в создании новых лекарственных препаратов является конструирование пептидомиметиков на базе структур эндогенных пептидов. С одной стороны, этот подход позволяет использовать плюсы пептидных соединений, обеспечивая минимальную токсичность и отсутствие побочных эффектов у конструируемых потенциальных препаратов. С другой стороны, использование структур, лишь имитирующих пептиды, избавляет от минусов, свойственных эндогенным соединениям, то есть ферментативной нестабильности и многофункциональности.
Целью настоящего исследования является создание на базе структуры ХЦК-4 нового эффективного потенциального анксиолитика, свободного от побочных эффектов, характерных для транквилизаторов бензодиазепинового ряда и не уступающего им по эффективности.
В соответствии с поставленной целью в данной работе решались следующие задачи:
— конструирование на базе ХЦК-4 новых пептидомиметиков с потенциально анксиолитическими свойствами и их синтез;
— изучение взаимосвязи «структура-активность» в ряду синтезированных соединений и отбор наиболее перспективного потенциального анксиолитика;
— изучение биологически активной конформации синтезированных аналогов ХЦК-4;
— синтез ряда конформационно ограниченных соединений на базе аналогов ХЦК-4;
— оптимизация синтеза наиболее активного соединения, отобранного для развития в качестве лекарственного средства.
Выводы.
1. Разработан подход к дизайну пептидных антагонистов пептидных рецепторов, являющийся развитием топохимического принципа.
2. Сконструирована и синтезирована новая группа И-ацилдипептидных триптофансодержащих аналогов холецистокинипа-4 с анкиолитической или анксиогенной активностью.
3. Изучена связь структуры и активности в ряду полученных аналогов холецистокинина. Показано, что соединения на основе 1)-триптофана обладают анксиогенной, а соединения на основе ¿—триптофана — анксиолитической активностью. Показано, что основными фармакофорными элементами являются фенильное и индолильное ароматические ядра. Выявлена зависимость активности от расстояния между ароматическими фармакофорами: оптимальным является расстояние в 13 а-связей, равное таковому между этими фармакофорами в природном ХЦК-4.
4. С помогцью 'Н-ЯМР-спектроскопии изучена зависимость активности от предпочтительной конформации И-ацилдипептидного аналога в растворе. Аналоги с предпочтительной (3-поворотной структурой активны, соединения с у-поворотной структурой неактивны.
5. С использованием конформационно ограниченных соединений показано, что наиболее вероятно биологически активной конформацией И-ацилдипептидных ретро-аналогов ХЦК-4 является (3-поворотная структура П-го типа.
6. С использованием компьютерного моделирования показано фармакофорное подобие дипептидных аналогов с анксиогенной активностью и природного ХЦК-4.
7. Наиболее активный ]Ч-ацилдипептидный аналог ХЦК-4, амид N-(6-фенилгексаноил)глицил-/,-триптофана (ГБ-115), отобран в качестве перспективного селективного анксиолитика и в настоящее время находится на завершающей стадии доклинических исследований.
Заключение
.
Таким образом, в данной работе впервые сконструированы и синтезированы Ы-ацилзамещённые дипептидные аналоги холецистокинина-4 с анксиолитической/анксиогенной активностью. Наиболее активный из них отобран для развития в качестве потенциального анксиолитического препарата. Методами 'Н ЯМР-спектроскопии в растворе и с помощью конформационно-ограниченных аналогов получены данные о том, что биологически активной конформацией этих соединений является р-изгиб П-го типа.
